重组草酸脱羧酶的表达及诱导条件优化

Expression and Optimized Inducing Conditions of Recombinant Oxalate Decarboxylase

摘要: 本文研究了枯草芽胞杆菌(Bacillus subtilis)来源的重组草酸脱羧酶的诱导表达,并对诱导条件进行了优化。结果表明,重组草酸脱羧酶的最佳诱导表达条件为:发酵液初始pH 8.0,诱导时机为3 h,添加IPTG 0.4 mmol/L,MnCl2 6 mmol/L,诱导表达10 h,在此条件下每升发酵液产草酸脱羧酶960 U。

Abstract: The optimized inducing condition of recombinant oxalate decarboxylase derived from Bacillus subtilis was studied. The results showed that 960 U oxalate decarboxylase per liter was induced under the conditions as follows: initial pH at 8.0, the recombinant strain was induced by adding 0.4 mmol/L IPTG and 6 mmol/L MnCl2 after 3 h, and cultured for 10 h.
