补硒对血糖受损大鼠胰岛素抵抗和胰岛素敏感性的影响

Effect of Selenium Supplementation on Insulin Resistance and Insulin Sensitivity in Rats with Impired Fsting Glucose

摘要: 为了探讨补硒对空腹血糖受损大鼠血糖、胰岛素水平、胰岛素抵抗及胰岛素敏感性的影响,将40只大鼠随机分成4组,对照组、单纯高脂饲料组(HFD,饲料硒含量0.3 mg/kg),另外两组在HFD基础上添加富硒小麦(HFD+Se+,硒含量0.6 mg/kg;HFD+Se++,硒含量1.2 mg/kg)。实验前和实验第5周、10周、16周采血测定空腹血糖(FBG)和空腹胰岛素(FINS),计算胰岛素抵抗指数(HOMA-IR)和胰岛素敏感指数(ISI)。结果发现,无论补硒与否,高脂喂养大鼠体重明显高于对照组。从第10周开始3个高脂组FBG和FINS明显上升,提示空腹血糖受损。第16周时,HFD+Se+组和HFD+Se++组FINS显著低于HFD组,HFD+Se+和HFD+Se++组HOMA-IR显著低于HFD组,HFD+Se++组ISI显著高于HFD组。结果说明,补硒改善了空腹血糖受损大鼠的胰岛素抵抗,提高了胰岛素敏感性。

Abstract: To investigate the effects of selenium (Se) supplementation on fsting blood glucose (FBG) level, fsting insulin (FINS) level, insulin resistance and insulin sensitivity in rats with impired fsting glucose, 40 rats were randomly divided into control group, high-fat diet (HFD, 0.3 mg/kg Se) group, HFD supplemented with two levels of Se (HFD+Se+, 0.6 mg/kg Se; HFD+Se++, 1.2 mg/kg Se) groups. FBG and FINS were measured at the beginning and the fifth, tenth, sixteenth weeks of the experiment. Insulin resistance index (HOMA-IR) and insulin sensitivity index (ISI) were calculated according FBG and FINS. As a result, the body weight of rats was significantly higher in high-fat diet groups (including Se supplementation) than control group. From the tenth week, FBG and FINS significantly increased in 3 groups with high fat diet, demonstrating impaired fasting glucose in these rats. At sixteenth weeks, FINS was significantly decreased in HFD+Se+ and HFD+Se++ groups than in HFD group. HOMA-IR was also significantly decreased in HFD+Se+ and HFD+Se++ groups than in HFD group. On the other hand, ISI was significantly increased in HFD+Se++ group than in HFD group. In conclusion, Se supplementation improves insulin resistance and increases insulin sensitivity.
