湿地环境可培养粘细菌多样性研究

Study on Diversity of Cultivable Myxobacteria in Wetland Environment

摘要: 采用辅助菌诱导法和滤纸诱导法对广东不同类型湿地底泥样品进行粘细菌的分离纯化,并将形态鉴定与16S rDNA基因序列分析相结合,对纯培养物进行初步分类鉴定。从84份样品中共分离纯化到67株粘细菌,初步鉴定为:粘球菌属(Myxococcus)32 株、珊瑚球菌属(Corallococcus)15株、孢囊杆菌属(Cystobacter)2株、多囊菌属(Polyangium)5株、原囊菌属(Archangium)7株、匣状球菌属(Pyxidicoccus)5株、波管状菌属(Hyalangium)1株。结果表明,湿地环境粘细菌多样性丰富,并且存在大量的新类群。研究结果为湿地环境粘细菌资源的挖掘提供了理论支持。

Abstract: Purpose of the work was to isolate myxobacteria from different types of wetland sediment in Guangdong by help-bacteria and filter papers methods. These strains were preliminary identified based on the morphological characteristics and the analysis of 16S rDNA gene sequences. 67 strains were isolated from 84 samples. The pure cultures could be classified into 7 genera: Myxococcus (32 strains), Corallococcus (15 strains), Cystobacter (2 strains), Polyangium (5 strains), Archangium (7 strains), Pyxidicoccus (5 strains) and Hyalangium (1 strain). The results showed that myxobacteria were widely distributed in wetland environment, and lots of uncultured myxobacteria resources existed in this region. This work laid a foundation for further researches on the exploitation of myxobacteria in wetland environment.
