大孔树脂对红薯叶总黄酮的吸附及解吸特性研究

Absorption and Desorption of Total Flavonoids from Sweet Potato Leaves with Macroporous Resins

摘要: 以红薯叶为原料,选用D101、AB-8、DM130大孔吸附树脂进行静态吸附及解 吸试验,筛选出适合吸附分离红薯叶总黄酮的树脂并确立其最佳吸附及解吸工艺参 数。试验结果表明,AB\|8大孔吸附树脂对红薯叶总黄酮吸附、解吸效果较好,最佳 吸附条件为上样液浓度0.3 mg/mL,上样液流速2 mL/min,上样液pH 5.0,吸附率为 66.8%;最佳解吸条件为洗脱液浓度90%,洗脱液流速4 mL/min,洗脱液用量5 BV, 解吸率为70.3%。

Abstract: Static adsorption and desorption capacity were assayed to screen the proper macroporous resin for total flavonoids from sweet potato leaves, and most suitable manipulation parameters of macroporous resin were confirmed. The results showed that AB-8 macroporous resin had strong adsorption and desorption capacity among the three resins used in this experiment. The most appropriate adsorption conditions were: sample concentration of 0.3 mg/mL, sample flow velocity of 2 mL/min, pH 5.0, and the adsorption rate reached 66.8%. The most fitting elution conditions were: ethanol concentration of 90%, elution rate of 4 mL/min, elution volume 5 BV, and the desorption rate was 70.3%.
