组培技术提高白背三七试管苗中总黄酮含量

The Improvement of Total Flavonoids Content of Gynura divaricata Callus by the Tissue Culture Technology

摘要: 为了建立一种提高白背三七试管苗总黄酮含量的组织培养技术,采用组织培养技术,改变激素配比、碳源、微量元素(Mn2+:Zn2+)比例和氮源,来提高白背三七试管苗中总黄酮的含量。正交实验结果表明,当碳源为蔗糖30 g/L、微量元素Mn2+:Zn2+比为44.6 mg/L:13.2 mg/L、氮源KNO3:NH4NO3 为2 020 mg/L:1 600 mg/L、激素比NAA:6-BA为0.1 mg/L:1.5 mg/L时,白背三七试管苗中总黄酮含量相对于原盆栽植株提高了15%。通过组织培养技术建立了一种可提高白背三七总黄酮含量的繁殖体系,为白背三七优良种苗的培育提供技术支持,该研究结果也表明利用组织培养技术改变培养基中物质配比有益于植物生长和次生代谢产物积累。

Abstract: The test mainly aimed at the establishment of a tissue culture technology to improve the total flavonoids of Gynura divaricate callus. The tissue culture technology was used to change the MS medium with different hormone combination, carbon source, nitrogen source and trace element ratio. The orthogonal experiment results showed that the maximum total flavones content was increased by 15% when the NAA∶6-BA was 0.1 mg/L∶1.5 mg/L, carbon source was 30 g/L, Mn2+∶Zn2+ was 44.6 mg/L∶13.2 mg/L and nitrogen source KNO3∶NH4NO3 was 2 020 mg/L∶1 600 mg/L. A system of tissue culture was developed for the enhancing of the total flavonoid of the G. divaricata DC, which was helpful for the seeding cultivation technology of G. divaricata DC and provided essential principles and techniques of improving the secondary metabolism for further research.
