Optimization of Pentostatin Fermentation Conditions

摘要: 喷司他丁是一种抗肿瘤药物,主要以微生物发酵法生产。本研究对喷司他丁发酵的通气量、培养温度、接种量、种龄、发酵周期等参数进行单因素考察,并通过正交试验确定了喷司他丁最佳发酵参数为:装量60 mL,种龄42 h,按5%接种量,28℃下发酵168 h,喷司他丁发酵效价达183 μg/mL,比对照提高20%以上。

Abstract: Pentostatin is an anticancer drug, mainly produced by microbial fermentation. In this study, fermentation ventilation, temperature, inoculum, seed age, and other parameters of the fermentation cycle were discussed, then a orthogonal experiment was done to determine the optimum fermentation parameters: loading volume of 60 mL, seed age of 42 h, 5% inoculum, 28℃ for 168 h fermentation, pentostatin fermentation titer reached 183 μg/mL, which was increased by 20% compared to the control.
