高效液相色谱法测定转化液中海藻糖

Determination of Trehalose in Transformation Liquid by High Performance Liquid Chromatography

摘要: 以麦芽糖为反应底物,与从土壤中筛选的产海藻糖合酶菌株发酵制得的粗酶液反应制取转化液。采用高效液相色谱法以Sugar-PakTM Ⅰ为色谱柱测定转化液中海藻糖的含量。研究结果显示,该方法能够将海藻糖和葡萄糖完全分开,与DNS比色法所得结果一致,为1.5 g/100mL,但HPLC法在测定海藻糖含量时,操作过程及数据处理更为简单,易于掌握。

Abstract:

Transformation liquid was obtained from the reaction of crude enyme with maltose as oligomer. The crude enyme was fermented by trehalose synthase producing strain which was selected from soil. The concentration of trehalose in the transformation was determined by HPLC with Sugar-PakTM Ⅰ. Results showed that the method could separate the trehalose and glucose completely, accordance with the result of DNS colorimetric method of 1.5g/100mL, but the HPLC operation process and data processing were simpler and easier to control in trehalose content determination.
