Determination of Raspberry Ketone in Raspberry by HPLC-MS/MS

摘要: 建立了树莓中树莓酮含量检测的高效液相色谱-串联质谱(HPLC-MS/MS)分 析方法。本研究采用甲醇作为提取溶剂热回流提取树莓酮,通过化学方法除去粗提 物中杂质并分离出其中的醛酮等羰基化合物,色谱分离后进行HPLC-MS/MS多反应监 测模式下的定性及定量分析。结果显示,不同基质中树莓酮的定量检测限为1.0 μg/kg,回收率为754%~82.3%,最大相对标准偏差为8.40%。本方法分析速度快 ,灵敏度高,重现性好,可用于树莓中树莓酮的检测。

Abstract: A high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) method was developed for analysis of raspberry ketone in raspberry. Raspberry ketone was extracted by hot refluxing method with methanol as extraction solvent. The impurities in crude extract were removed by chemical method, and the aldehyde, ketone and other carbonyl compounds were isolated from crude extract. Qualitative and quantitative analysis was carried out for the analyte under the MRM mode after the chromatographic separation. The quantitative limit of detection of RK in different matrix was 1.0 μg/kg. The average recovery was 75.4%~82.3%, and the relative standard deviation was 5.97%~8.40%. This method is accurate,rapid,sensitive,and adapt to the inspection of the raspberry ketone in raspberry.
