超滤耦合径向流色谱分离纯化灵芝多糖的研究

Purification of Ganoderma lucidum Polysaccharides by Coupling Ultrafiltration with Radial Flow Chromatography

摘要: 研究了超滤及其径向流耦合对灵芝多糖酶解液中蛋白质的脱除效果及多糖截 留率。试验选择截留分子量为100 kDa的超滤膜,研究了料液体积流量、超滤时间、 浓缩倍数对多糖超滤效果的影响;比较了超滤耦合径向流色谱法、单一色谱法和 Sevag法的脱蛋白效率。结果表明,超滤法脱除蛋白质的适宜工艺参数为:室温、压 力小于30 psi,料液体积流量100 mL/min,超滤时间60 min,浓缩倍数15~17倍, 此条件下蛋白质去除率为82.21%,多糖截留率为63.32%。将超滤与径向流色谱耦合 ,蛋白质脱除率及多糖截留率分别高达99.24%和89.52%,相比单一色谱法及Sevag法 ,其效率分别明显提高。该项研究表明,超滤耦合径向流色谱脱除蛋白质是可行的 ,并具有较高的多糖截留率。

Abstract: The removal of protein and the retention rate of polysaccharide in Ganoderma lucidum polysaccharides hydrolyzate were studied using ultrafiltration and coupling method. Using the ultrafiltration membrane with a cut-off molecular mass of 100 kDa, the effects of several parameters such as the flow rate, ultrafiltration time and concentration multiple on the ultrafiltration were evalutated. The optical conditions of removing protein by ultrafiltrating were determined. It is founded that the flow rate, time and concentration multiple were 100 mL/min, 60 min,15~17 times, respectively, at the room temperature and the pressure less than 30 psi. The rate of protein removal reached 82.21% and the retention rate of polysaccharide was 63.32%. Coupled the ultrafiltration with radial flow chromatography, the retention rate of polysaccharide and the removal rate of protein were as high as 99.24% and 89.52%. As compared with the single chromatography and the Sevag method, the efficiency increased. These findings thus clearly suggested purification of polysaccharides from Ganoderma lucidum by coupling ultrafiltration with radial flow chromatography was feasible with high retention rate.
