抗糖尿病药物治疗阿尔茨海默病的研究进展

摘要: 阿尔茨海默病(Alzheimers disease, AD)是以认知缺陷为主要特征的慢性疾病,目前尚无有效根治药物。由于患者数量显著增长,探究治疗AD的药物成为国内外的研究热点。近年流行病学研究表明,2型糖尿病是AD的危险因素,两者具有共同的病理生理机制,如胰岛素抵抗、淀粉样蛋白沉积、Tau蛋白过度磷酸化、炎症反应和氧化应激等。因此,从现有的抗糖尿病药物中筛选AD的治疗药物是目前研究的一种趋势。越来越多的研究已证实降糖药物(如胰岛素、二甲双胍等)具有改善AD病变的有益作用。从AD与2型糖尿病的相关性、抗糖尿病药物治疗AD两个方面综述了抗糖尿病药物治疗AD的研究进展,以期为AD的治疗拓宽思路。

Abstract: Alzheimers disease (AD) is a chronic disease characterized by cognitive deficiency. Currently, there is no effective cure drug for AD. Due to the significant increase in the number of patients, the exploration of drugs for the treatment of AD has become a research hotspot at home and abroad. In recent years, epidemiological studies have shown that type 2 diabetes mellitus is a risk factor for AD, and both of them share common pathophysiological mechanisms, such as insulin resistance, amyloid-β protein deposition, Tau hyperphosphorylation, inflammatory response and oxidative stress. Therefore, screening drugs for AD from existing anti-diabetic drugs is a trend of current research. More and more studies have confirmed that hypoglycemic drugs (such as insulin, metformin, etc.) have a beneficial effect on improving AD lesions. In this paper,  we summerized the progress of treatment of AD with antidiabetes drugs. The correlation between AD and type 2 diabetes mellitus and the treatment of AD with anti-diabetic drugs was also reviewed, in order to broaden the ideas for the treatment of AD.
