响应面分析法优化酶法提取红景天苷的工艺研究

Optimization of Enzymatic Extraction Process of Salidroside by Response Surface Analysis

摘要:

为研究纤维素酶酶解法提取大花红景天中红景天苷的最佳工艺,在单因素和正交实验数据的基础上,以红景天苷得率为响应值,进行响应面分析设计,考察酶用量、酶解温度、酶解pH对红景天苷得率的影响。结果显示,最佳提取工艺条件为:纤维素酶用量为药材重量的0.36%,酶解温度45 ℃、酶解pH 5.5,提取率为1.39%±0.31%。研究结果表明,该提取方法可减少酶用量,节能环保,有效提高红景天苷的提取率,可应用于大花红景天中红景天苷的提取。研究结果可为大花红景天中红景天苷产品的开发及应用提供科学参考。

Abstract:

The aim of this study was to research the optimum condition on extraction process of salidroside from Rhodiola crenulata with the enzymatic of cellulase method. The response surface methodology(RSM) was conducted based on single factor tests and orthogonal experiments. The effect of enzyme dosage, enzymolysis temperature and enzymolysis pH on extraction rate were tested. The optimal extraction condition were enzyme dosage 0.36%, extraction temperature 45 ℃, extraction pH 5.5, the extraction rate was 1.39%±0.31%. In summary, this energy-saving and environment-friendly extraction process could significantly reduce the amount of cellulase, and effectively improve the extraction yield of salidroside. Therefore, this process can be applied in the extraction of salidroside from Rhodiola crenulata. The results provided a scientific reference for developing and utilization of salidroside from Rhodiola crenulata.
