基于ITS序列分析对海南普通野生稻籼粳分化的研究

Study on Genetic Divergence of Hainan Common Wild Rice (Oraza rufipogon) Based on ITS Analysis

摘要: 对海南普通野生稻的18个居群分别进行ITS序列克隆和测序,研究海南普通野生稻居群的籼粳分化。结果表明,海南普通野生稻的ITS序列居群间差异性显著,ITS1长度为163~239 bp,G+C含量变化范围为53.5%~77-2%。ITS2长度为165~243 bp,G+C含量变化范围为54.5%~74.2%。ITS1和ITS2区简约信息位点分别为108个和145个,单一信息位点分别为7个和2个。InDel(插入/缺失)位点分别为133个和145个,发现了9个ITS籼粳特异性位点。ClustalX软件排序及Mega构树结果表明,海南普通野生稻存在籼粳分化,偏籼型有10个居群,偏粳型有8个居群。本研究有助于揭示海南普通野生稻在起源演化上的地位,并为有效利用海南优良野生稻种质资源提供理论依据。

Abstract: Eighteen individual samples of common wild rice (Oryza rufipogon Griff.) from Hainan were used for cloning and sequencing. Sequence analysis showed significant differences in ITS sequence among these populations. The results indicated that these populations had a genetic relationship, a total of 108 polymorphic fragments were detected in ITS1, and 145 polymorphic fragments in ITS2. Besides, there were 133 InDels in ITS1, 145 InDels in ITS2. It indicated that the common wild rice from Hainan performed some Indica-Japonica differentiations, more Indica than Japonica types, and 9 ITS Indica-Japonica differentiations sites were found. This work will help to understand the status of Oraza ruffipogon from Hainan in the origin and evolution of the cultivated rice in China, and also offer reference for potential use of the wild rice from Hainan.
