摘要:

为探讨长链非编码RNA锌指核转录因子反义链1(zinc finger nuclear transcription factor antisense RNA 1,ZFAS1)在结直肠癌中的表达及生物学功能,通过实时荧光定量PCR检测ZFAS1在结直肠癌中的表达水平,并分析其表达水平与患者临床病理特征之间的关系。利用细胞功能学试验检测ZFAS1对结肠癌细胞生长及迁移能力的影响,敲低ZFAS1,检测其潜在靶基因锌指核转录因子1(zinc finger nuclear transcription factor X-box binding 1-type containing 1,ZNFX1)在结肠癌细胞系中的表达量,并通过细胞功能学试验检测ZNFX1对结肠癌细胞生长及迁移能力的影响。结果显示,ZFAS1在结直肠癌细胞中高表达,敲低ZFAS1可以抑制结肠癌细胞的生长及迁移;敲低ZFAS1后,ZNFX1的表达上调;ZNFX1在结肠癌细胞系中低表达,过表达ZNFX1可以抑制结肠癌细胞的生长及迁移能力。研究结果为阐明ZFAS1在结直肠癌中的作用机制奠定了一定的理论基础。

Abstract:

To explore the expression and biological function of long noncoding RNA (lncRNA) zinc finger nuclear transcription factor antisense RNA 1 (ZFAS1) in colorectal cancer, the expression level of ZFAS1 in colorectal cancer were detected by real-time PCR, and the relationship between its expression level and clinicopathological features was analyzed. The effect of ZFAS1 on the growth and migration of colorectal cancer cells was detected by cell function experiment. By knockdown of ZFAS1, the expression of its potential target gene zinc finger nuclear transcription factor 1 (ZNFX1) in colorectal cancer cell lines were delected. The effect of ZNFX1 on the growth and migration of colon cancer cells was detected by cell function experiment. The results showed that, ZFAS1 was highly expressed in colorectal cancer, and knockdown of ZFAS1 could inhibit the growth and migration of colorectal cancer cells. After knockdown of ZFAS1, ZNFX1 expression was up-regulated. ZNFX1 was lowly expressed in colorectal cancer cell lines. Overexpression of ZNFX1 could inhibit the growth and migration of colorectal cancer cells. The results of this study laid a theoretical foundation for clarifying the mechanism of ZFAS1 in colorectal cancer.
