腹腔注射乙酸钠对斑马鱼食欲调控基因及胰岛素分泌基因的影响

Effect of Intraperitoneal Injection of Sodium Acetate on Zebrafish Appetite Regulatory Genes and Insulin Secretion Genes

摘要: 肠道微生物菌群发酵体内不易消化的碳水化合物可以产生乙酸。哺乳动物中,乙酸被报道可增加宿主食欲,改善葡萄糖平衡以及胰岛素敏感性。然而水产动物中乙酸盐相关作用及机理研究较少。以斑马鱼为研究对象,腹腔注射不同浓度乙酸钠发现,斑马鱼中促进摄食的基因表达量显著升高,而对抑制摄食基因的表达量没有显著影响,说明乙酸钠能够提高斑马鱼的食欲。另一方面,研究发现腹腔注射斑马鱼2 h、4 h、6 h后,肝脏及肠道促进胰岛素分泌基因表达水平也有显著性上升,说明乙酸钠对于胰岛素分泌具有促进作用。研究结果表明在水产动物中乙酸钠能够提高食欲并促进胰岛素的分泌,研究结果阐述了乙酸盐对水产动物生长的作用机理,以期为乙酸钠在生产中的应用提供依据。

Abstract: Acetic is produced from carbohydrates which are not digested by the intestinal microflora in the body. In mammals, acetic acid has been reported to increase the appetite of the host, improved glucose balance and insulin sensitivity. However, there are few studies on the mechanism of acetate in aquatic animals. In this experiment, zebrafish was used as the research object. Intraperitoneal injection of different concentrations of sodium acetate in zebrafish promoted the expression of orexigenic gene, but had no significant effect on the anorexigenic genes, indicating that sodium acetate can improve zebrafish appetite. On the other hand, the study found that intraperitoneal injection of sodium acetate promoted the expression of insulin secretion enhancement genes in the liver and intestine of zebrafish at 2 h, 4 h and 6 h post injection, indicating that sodium acetate has a positive effect on insulin secretion. Therefore, our research showed that sodium acetate can improve the appetite and promote the secretion of insulin in aquatic animals, expounded the mechanism of acetic acid on the growth of aquatic animals, and provided basis for the application of sodium acetate in production.
